The editor
This special issue reports the results of pharmacologic and biochemical studies evaluating the mode of action and applications of nomifensin, a chemically new substance which shows promise in the treatment of depression. In 9 contributions from internationally recognized researchers, the work explores virtually all topics relevant to the development of this new antidepressant. The value of this work is further extended through details of innovative methodological strategies and findings that contribute to increased understanding of the biochemistry of depression.

Selected studies featured in this issue
- a pharmacokinetic/pharmacodynamic study demonstrating that serum kinetics are not necessarily directly related to a drug's CNS effects: with nomifensin, CNS effects assessed by the computer EEG last much longer than serum levels would indicate
- an investigation, using a sensitive experimentál paradigm, of anticholinergic effects which have been a common problem plaguing tricyclic antidepressants: among the whole spectrum of antidepressants studied, nomifensin was found to belong to the group with minimai anticholinergic effects
- double-blind studies comparing nomifensin with other antidepressants and with placebo to determine therapeutic efficacy: in all cases, nomifensin proved to be therapeutically equivalent or superior
- a double-blind study evaluating long-term treatment using nomifensin on the one hand, and imipramine on the other: while initial improvement was impressive, relapses occurred with both drugs during one year of treatment.
